RESUMEN

There is an increased awareness of the importance of data publication, data sharing, and open science to support research, monitoring and control of vector-borne disease (VBD). Here we describe the efforts of the Global Biodiversity Information Facility (GBIF) as well as the World Health Special Programme on Research and Training in Diseases of Poverty (TDR) to promote publication of data related to vectors of diseases. In 2020, a GBIF task group of experts was formed to provide advice and support efforts aimed at enhancing the coverage and accessibility of data on vectors of human diseases within GBIF. Various strategies, such as organizing training courses and publishing data papers, were used to increase this content. This editorial introduces the outcome of a second call for data papers partnered by the TDR, GBIF and GigaScience Press in the journal GigaByte. Biodiversity and infectious diseases are linked in complex ways. These links can involve changes from the microorganism level to that of the habitat, and there are many ways in which these factors interact to affect human health. One way to tackle disease control and possibly elimination, is to provide stakeholders with access to a wide range of data shared under the FAIR principles, so it is possible to support early detection, analyses and evaluation, and to promote policy improvements and/or development.

A spatiotemporal investigation of hematophagous fly prevalence was conducted over a 1-year period on 12 beef cattle farms located in major livestock areas of Bangkok, Thailand, using Vavoua traps. The survey revealed 5,018 hematophagous flies belonging to Muscidae and Tabanidae, with the 3 dominant species identified as Stomoxys calcitrans (Linnaeus) (2,354; 46.91%), Musca crassirostris Stein (1,528; 30.45%), and Haematobia exigua de Meijere (922; 18.37%). The abundance of S. calcitrans per trap per week was significantly higher during the rainy season (45.64 ±â€…14.10), followed by the cold and dry seasons (6.39 ±â€…2.16 and 3.04 ±â€…1.27, respectively). The relative abundance of S. calcitrans reached the highest apparent density per trap per day (ADT) index of 9.83 in August 2022 during the rainy season. Subsequently, there was a rapid decline, and the ADT index dropped to nearly zero in December 2022 during the cold season. This low abundance continued through the dry months from March to May 2023. The higher rainfall and relative humidity could significantly contribute to the high relative abundance of S. calcitrans. In contrast, M. crassirostris and H. exigua showed population fluctuations that were not significantly associated with seasonal changes and weather conditions. Remote sensing data and spatial regression analyses using ordinary least squares regression showed the high spatial density of S. calcitrans in the north direction of the Khlong Sam Wa district during the rainy season; it shifted toward the south in the cold and dry seasons, corresponding with rainfall.

The mosquito Aedes aegypti, known to transmit important arboviral diseases, including dengue, chikungunya, Zika and yellow fever. Given the importance of this disease vector, a number of control programs have been proposed involving the use of the sterile insect technique (SIT). However, the success of this technique hinges on having a good understanding of the biology and behavior of the male mosquito. Behavioral responses of Ae. aegypti male populations developed for SIT technology were tested under laboratory conditions against chemical and natural irritants and repellents using an excito-repellency (ER) chamber. The results showed that there were no significant behavioral escape responses in any of the radiation-sterilized male Ae. aegypti test populations when exposed to citronella, DEET, transfluthrin, and deltamethrin, suggesting that SIT did not suppress the expected irritancy and repellency (avoidance) behaviors. The type of information reported in the current study is vital in defining the effects of SIT on vector behavior and understanding how such behavior may influence the success of SIT technology with regard to other vector control interventions.

The growth and associated traits of Aedes aegypti (L.) mosquitoes may adapt and evolve in response to the costs associated with body size in relation to latitudinal variation. We analyzed the life-history traits and energy reserves of field-collected mosquitoes from Taiwan, Thailand, and Indonesia along a latitudinal range spanning from 23°N to 6°S. A U-shaped relationship between body size and latitude was observed. Our study demonstrated the role of latitudinal temperature variations in determining the body size patterns of Ae. aegypti. Notably, the body size of the northern populations (from Taiwan) was significantly larger than those of the tropical populations from Thailand and Indonesia. Models have demonstrated that regional precipitation levels may contribute to body trait variations in certain high-latitude populations in Thailand. However, Indonesian populations have high development rates and large body sizes, indicating the involvement of other physiological traits in determining mosquito body size. The reproductive output of the adult females in this study was positively correlated with body size, but our measure of longevity did not covary significantly with the body size. By contrast, the reproductive output of mosquito-tested populations was in inverse proportion to longevity. Additionally, the mean teneral glycogen levels in the Indonesian and Thai populations were 2.5 times higher than those in the Taiwanese populations. The Indonesian and Thai populations had low mean generation and doubling times, resulting in a high intrinsic rate of increase compared with that of the Taiwanese populations, despite the Taiwanese populations having the highest net reproduction rate.

WHO tube and CDC bottle bioassays are currently available for insecticide resistance monitoring and malaria transmission research. Multiple parameters including mosquito density, age, and nutritional status may affect the readout in these bioassays' tests. This study aims to assess the effects of experimental factors on knockdown and mortality measurements in dominant malaria vectors in Thailand following exposure to sublethal and lethal doses of transfluthrin. The effects of (i) 3 different mosquito batch sizes (5, 10, and 20 individuals) and (ii) 2 age groups (3-5 and 20-23 days old) on outcomes measured using the WHO tube (14.7 µg/cm2) and CDC bottle bioassay discriminating concentration (0.006 µg/cm2) against 2 laboratory strains: Anopheles dirus Peyton & Harrison and Anopheles minimus Theobald (species A) and wild-caught Anopheles harrisoni Harbach & Manguin (species C). Our results showed higher knockdown at 1-h exposure using WHO tube and CDC bottle bioassays containing 20 individuals compared to batches containing 10 and 5 individuals. Older mosquitoes showed greater susceptibility than younger test population, especially for An. mininus. Our study supports WHO recommendations for using 3- to 5-day-old mosquitoes. It also validates Praulin et al. (2022) proposal to divide the cohort into smaller batches with more test replicates when it is not practicable to test 25 mosquitoes per replicate.

Volatile pyrethroids exert a range of both lethal and behavioral effects on mosquitoes through the passive release of insecticides into the atmosphere. We investigated the protective efficacy (PE) of transfluthrin-treated jute (TI-jute) and cotton (TI-cotton) fabrics, worn at the back of a protective black vest, against laboratory-reared pyrethroid susceptible and resistant strains of Aedes aegypti (L.) in a semifield system (SFS). Each fabric (1,029 cm2) was treated with 1.79 mg/cm2 of transfluthrin as the intervention. Human landing collections were conducted by 2 collectors seated in designated treatment and control compartments of the SFS. The trials were conducted for 41 days, with 16 days partitioned into morning and evening phases. Furthermore, we examined blood feeding behavior and fecundity of the surviving mosquitoes post-exposure. Results showed that in the morning, the PE of TI-jute (49.4%) was higher than that of TI-cotton (36.8%). TI-jute demonstrated a lower PE of 9.6% against the transfluthrin-resistant strain. Remarkably, a significantly higher number of eggs were laid by the transfluthrin-resistant mosquitoes that survived the intervention (36.5 eggs/female) compared to the control group (11.8 eggs/female). These findings suggest that TI-jute can help protect against bites and alter the life traits of Ae. aegypti. The study highlights that the timing of the intervention during the day affected the efficacy of TI-jute and TI-cotton, while sublethal exposure to transfluthrin stimulated egg production in the resistant strain. These are critical challenges that warrant attention in vector control strategies. Investigating this phenomenon in mosquito reproduction necessitates future research at a molecular level.

The effectiveness of vector-control tools is often assessed by experiments as a reduction in mosquito landings using human landing catches (HLCs). However, HLCs alone only quantify a single characteristic and therefore do not provide information on the overall impacts of the intervention product. Using data from a recent semi-field study which used time-stratified HLCs, aspiration of non-landing mosquitoes, and blood feeding, we suggest a Bayesian inference approach for fitting such data to a stochastic model. This model considers both personal protection, through a reduction in biting, and community protection, from mosquito mortality and disarming (prolonged inhibition of blood feeding). Parameter estimates are then used to predict the reduction of vectorial capacity induced by etofenpox-treated clothing, picaridin topical repellents, transfluthrin spatial repellents and metofluthrin spatial repellents, as well as combined interventions for Plasmodium falciparum malaria in Anopleles minimus. Overall, all interventions had both personal and community effects, preventing biting and killing or disarming mosquitoes. This led to large estimated reductions in the vectorial capacity, with substantial impact even at low coverage. As the interventions aged, fewer mosquitoes were killed; however the impact of some interventions changed from killing to disarming mosquitoes. Overall, this inference method allows for additional modes of action, rather than just reduction in biting, to be parameterised and highlights the tools assessed as promising malaria interventions.

Over the past two decades, there has been a worldwide resurgence in the bed bugs Cimex lectularius L. and Cimex hemipterus (F.). This is primarily due to insecticide resistance, making bed bug management and eradication challenging and expensive. To address the need for more affordable control solutions, "lethal harborages" were explored. Cardboard squares were treated using insecticidal dusts at different dosage levels, including silica dioxide, diatomaceous earth, deltamethrin, permethrin, and fipronil. Two strains of C. lectularius, one susceptible and one resistant, were allowed to enter the treated harborages, and mortality rates were recorded daily. The silica dioxide products proved to be the most efficacious, consistently achieving 100% mortality between 14-17 d at the highest dose. An artificial environment trial using the "new ChinChex®" formulation of silica dioxide resulted in the complete elimination of bed bugs in the treated harborages within 21 d. These findings suggest that lethal harborages, especially those impregnated with silica dioxide, offer a cost-effective solution that could be incorporated into broader integrated bed bug management strategies. This approach may help alleviate the burden of bed bug infestations in economically disadvantaged communities.

Mosquitoes play a crucial role as primary vectors for various infectious diseases in Thailand. Therefore, accurate distribution information is vital for effectively combating and better controlling mosquito-borne diseases. Here, we present a curated dataset of the mosquito distribution in Thailand comprising 12,278 records of at least 117 mosquito species (Diptera: Culicidae). The main genera included in the dataset are Aedes, Anopheles, Armigeres, Culex, and Mansonia. From 2007 to 2023, data were collected through routine mosquito surveillance and research projects from 1,725 locations across 66 (out of 77) Thai provinces. The majority of the data were extracted from a Thai database of the Thailand Malaria Elimination Program. To facilitate broader access to mosquito-related data and support further exploration of the Thai mosquito fauna, the data were translated into English. Our dataset has been published in the Global Biodiversity Information Facility, making it available for researchers worldwide.

Aedes aegypti is a major vector of dengue fever in tropical regions. Spatial repellents (SRs) have shown promise in delaying pesticide resistance. Methods for discriminating concentrations (DCs) are well established using various bioassay tools, while data for high-throughput screening system (HITSS) toxicity bioassay (TOX) are absent. In this study, we compared and optimized lethal (LCs) and sub-lethal concentrations (SLCs) of transfluthrin (TFT) and metofluthrin (MFT) on pyrethroid-susceptible (USDA) and pyrethroid-resistant (Pu-Teuy) Ae. aegypti (L.) strains, using the HITSS-TOX. Mean mortality (MT) was 100% at LC99 and DC, compared to LC50 (45.0 ± 3.7%) and LC75 (65.8 ± 7.0%) for the USDA strain. However, the resistant strain (Pu-Teuy) showed reduced susceptibility against TFT and a significantly lower MT at LC50 (12.5 ± 4.4%; t = 5.665, df = 10, p < 0.001), LC75 (9.2 ± 3.5%; t = 4.844, df = 10, p = 0.001), LC99 (55.0 ± 9.9%; t = 4.538, df = 5, p = 0.006), and DC (75.0 ± 5.2%; U = 3.0, p = 0.007). The DC of TFT (0.15222%) was 4.7-fold higher than for MFT (0.03242%) in USDA strain. The baseline DCs established are useful to better understand susceptibility and the efficacy of various repellents against field populations of Ae. aegypti.

Repellents play a major role in reducing the risk of mosquito-borne diseases by preventing mosquito bites. The present study evaluated the mosquito-repellent activity of ß-caryophyllene oxide 1% (BCO), vetiver oil 2.5% (VO), and their binary mixtures (BCO + VO (1:1), BCO + VO (2:1), BCO + VO (1:2)) against four laboratory-colonized mosquito species, Aedes aegypti (L.), Aedes albopictus (Skuse), Anopheles minimus Theobald, and Culex quinquefasciatus Say, using an excito-repellency assay system. In general, the compound mixtures produced a much stronger response in the mosquitoes than single compounds, regardless of the test conditions or species. The greatest synergetic effect was achieved with the combination of BCO + VO (1:2) in both contact and noncontact trials with An. minimus (74.07-78.18%) and Cx. quinquefasciatus (55.36-83.64%). Knockdown responses to the binary mixture of BCO + VO were observed for Ae. albopictus, An. minimus, and Cx. quinquefasciatus, in the range of 18.18-33.33%. The synergistic repellent activity of BCO and VO used in this study may support increased opportunities to develop safer alternatives to synthetic repellents for personal protection against mosquitoes.

BACKGROUND: The mosquito landing rate measured by human landing catches (HLC) is the conventional endpoint used to evaluate the impact of vector control interventions on human-vector exposure. Non-exposure based alternatives to the HLC are desirable to minimize the risk of accidental mosquito bites. One such alternative is the human-baited double net trap (HDN), but the estimated personal protection of interventions using the HDN has not been compared to the efficacy estimated using HLC. This semi-field study in Sai Yok District, Kanchanaburi Province, Thailand, evaluates the performance of the HLC and the HDN for estimating the effect on Anopheles minimus landing rates of two intervention types characterized by contrasting modes of action, a volatile pyrethroid spatial repellent (VSPR) and insecticide-treated clothing (ITC). METHODS: Two experiments to evaluate the protective efficacy of (1) a VPSR and (2) ITC, were performed. A block randomized cross-over design over 32 nights was carried out with both the HLC or HDN. Eight replicates per combination of collection method and intervention or control arm were conducted. For each replicate, 100 An. minimus were released and were collected for 6 h. The odds ratio (OR) of the released An. minimus mosquitoes landing in the intervention compared to the control arm was estimated using logistic regression, including collection method, treatment, and experimental day as fixed effects. RESULTS: For the VPSR, the protective efficacy was similar for the two methods: 99.3%, 95% CI (99.5-99.0) when measured by HLC, and 100% (100, Inf) when measured by HDN where no mosquitoes were caught (interaction test p = 0.99). For the ITC, the protective efficacy was 70% (60-77%) measured by HLC but there was no evidence of protection when measured by HDN [4% increase (15-27%)] (interaction test p < 0.001). CONCLUSIONS: Interactions between mosquitoes, bite prevention tools and the sampling method may impact the estimated intervention protective efficacy. Consequently, the sampling method must be considered when evaluating these interventions. The HDN is a valid alternative trapping method (relative to the HLC) for evaluating the impact of bite prevention methods that affect mosquito behaviour at a distance (e.g. VPSR), but not for interventions that operate through tarsal contact (e.g., ITC).

Aedes aegypti and Aedes albopictus (Aedes) transmit highly pathogenic viruses such as dengue, chikungunya, yellow fever, and Zika which can cause life-threatening diseases in humans. They are the most important vectors of arboviruses in Thailand. Their vectorial capacity (VC) is highly complex mainly due to the interplay between biotic and abiotic factors that vary in time and space. A literature survey was conducted to collate and discuss recent research regarding the influence of Aedes vector biology, behaviour, and ecology on arbovirus transmission in Thailand. The survey followed guidelines of preferred reporting items of systematic reviews and meta-analyses (PRISMA). All fields, keyword search was conducted in the Web of Science database for the period of 2000-2021. The search yielded 821 records on Ae. aegypti and 293 records on Aedes albopictus, of which 77 were selected for discussion. Genomic studies showed that there is a high genetic variation in Aedes albopictus whereas Ae. aegypti generally shows low genetic variation. Along with genetically unstable arboviruses, the interaction between Aedes and arboviruses is largely regulated by genomic events such as genetic mutations and immune response protein factors. Temperature and precipitation are the major climatic events driving arbovirus transmission. Human exposure risk factors are mainly due to multiple feeding patterns, including endophagy by Aedes albopictus and zoophagic behaviour of Ae. aegypti as well as diverse human-associated breeding sites. Integration of the One Health approach in control interventions is a priority with a rigorous focus on Aedes-arbovirus surveillance as a complementary strategy.

We evaluated the behavioral responses of Aedes albopictus and Anopheles minimus to 3 isovaleric acid and lactic acid-based chemical lure blends and 2 individual alcohols, using Spatial Repellency Assay in a high-throughput screening system (HITSS). Five doses of 0.0002, 0.001, 0.0025, 0.005, and 0.01 g were tested per lure. A BG-lure was used as a reference standard. After 10-min exposure, the number of mosquitoes moving toward or away from the treated chamber was calculated. The results showed that all lures were repellent against Ae. albopictus except for Lure-4 (4% w/v isovaleric acid + 2% w/v lactic acid + 0.0025% w/v myristic acid + 2.5% w/v ammonium hydroxide) which showed a nonsignificant attractancy at the lowest dose. Significantly high spatial repellency was observed at the highest dose of all the tested lures including BG-lure. Lure-2 (isoamyl alcohol) was significantly repellent at all the tested doses. Against An. minimus, Lure-5 (0.02% w/v isovaleric acid + 2% w/v lactic acid) showed significant spatial repellency while Lure-4 was significantly attractant, at all the tested doses. All lures, except Lure-4, showed strong spatial repellency at high doses and attractancy or weak spatial repellency at the lowest dose of 0.0002 g. In summary, our study demonstrated that spatial repellency and attractancy of the tested lures were influenced by both the dose tested and the mosquito species. Lure-2 and Lure-4 are potential spatial repellents and attractants, respectively, for malaria and dengue vectors. However, further studies are necessary to confirm these results at a semifield and open field level.

Haemoproteus columbae is a common haemosporidian parasite of wild pigeons (Columba livia) reported worldwide. In Thailand, the wild pigeon population is increasing due to paddy field monoculture. However, there are limited reports on the presence of H. columbae in these pigeon populations. The aim of the study was to characterize H. columbae in wild pigeons. A total of 87 wild pigeons were examined using microscopic and molecular methods. Haemoproteus columbae was detected in approximately 27.6% of pigeons and their morphological characteristics were described. The partial cytochrome b (cyt b) gene sequence of H. columbae was then characterized into three common lineages (HAECOL1, COLIV03, and COQUI05). By highlighting the morphologic and genetic characteristics of H. columbae commonly found in this population of pigeons, this study provides essential regional knowledge about haemosporidian parasites that could benefit future taxonomic and phylogeographic studies.

Complementary approaches to tackle outdoor and early evening biting mosquitoes are urgently required. Transfluthrin (TFT) is a volatile pyrethroid capable of altering mosquito feeding behavior. This study investigated the efficacy of TFT-treated jute (TFT-J) and cotton (TFT-C) fabrics on human landing activity, feeding and fecundity of Anopheles minimus in Thailand. Jute and cotton fabrics each measuring 1024 cm2 were impregnated with 7.34 × 10-4 g/cm2 TFT (20%, w/v), and evaluated in a semi-field screen house system. Two collectors, wearing an untreated control or TFT-treated vests, conducted human-landing collections of released 100 laboratory-reared adult females of An. minimus from 18:00-00:00 h for 16 consecutive nights. Recaptured mosquitoes were given a blood meal for 30 min. with a membrane feeding system for assessment of blood feeding and fecundity. TFT-J, relative to control, significantly reduced human landings (Odds Ratio (OR) =0.27 (95% Confidence Interval (CI) [0.10-0.74], p = 0.011)), however no significant reduction was observed for TFT-C (OR=0.67 [95% CI 0.24-1.82], p = 0.43). Blood feeding was significantly lower among mosquitoes exposed to TFT-J (12.45% [95% CI, 2.04-22.85], p = 0.029) and TFT-C (13% [95% CI, 0.99-26.84], p = 0.016) relative to control. Impregnated fabrics had no effect on the mean number of egg oviposition. However, egg hatchability was reduced in TFT-J (49.5% [95% CI, 21.74-77.26], p = 0.029) and TFT-C (40.2% [95% CI, 17.21-63.19], p = 0.008) relative to control. TFT-J significantly reduced the landing, blood feeding, and fertility of An. minimus. Further studies are needed to evaluate different treatment methods on fabrics and their incorporation in integrated mosquito management.

Each binary mixture formulation of Vetiveria zizanioides (L.) Nash (VZ) with Andrographis paniculata (Burm.f.) Wall. ex Nees (AP) or Cananga odorata (Lam.) Hook.f. & Thomson (CO) and AP with CO at 1:1, 1:2, 1:3, and 1:4 ratios (v:v) was investigated for behavioral responses on laboratory and field strains of Aedes aegypti. Irritant and repellent activities of each formulation were compared with N,N-diethyl-3-methylbenzamide (DEET) using an excito-repellency test system. The result demonstrated that the mixture of VZ:AP in all combination ratios was the most effective in inducing an irritancy response against the laboratory strain (56.57-73.33%). The highest percentage of escaped mosquitoes exposed to the mixture at a 1:4 ratio (73.33%) was significantly different from DEET (26.67%) (p < 0.05). Against the field strain, the strongest escape response of AP:CO at a 1:1 ratio in the contact trial (70.18%) was significantly different compared with DEET (38.33%) (p < 0.05). There was a weak non-contact escape pattern in all combinations of VZ:CO against the laboratory strains (6.67-31.67%). These findings could lead to the further development of VZ and AP as active ingredients in a repellent that could advance to human use trials.

The performances of the human-baited double net trap (HDNT) and the human-baited host decoy trap (HDT) methods were compared against the outdoor human landing catch (OHLC) method in Thailand and Vietnam. Two study sites were selected in each country: a rural village and a nearby forest setting. The three outdoor trap methods were rotated nightly between three set trapping positions, in a pre-assigned Latin square design. Volunteers were rotated following the trap rotation to avoid bias. The greatest number of adult mosquitoes was collected from the forest sites in both countries, showing Anopheles minimus (s.s.) Theobald (96.54%) and Anopheles dirus (s.s.) Peyton & Harrison (25.71%) as the primary malaria vectors in Thailand and Vietnam, respectively. At the Thai forest site, OHLC collected significantly more anopheline mosquitoes per trap night than HDNT and HDT, with mean ± standard error values of 14.17 ± 4.42, 4.83 ± 1.56, and 4.44 ± 1.45, respectively, whilst HDNT and HDT were significantly less productive at 0.34 times and 0.31 times, respectively, than OHLC in capturing anopheline mosquitoes. However, there were no significant differences among the three methods of trapping malaria vectors for the village site. At the Vietnamese forest site, HDNT achieved the highest performance in collecting Anopheline mosquitoes at 1.54 times compared to OHLC, but there was no significant difference between the two traps. The results suggested HDNT could be a possible alternative trap to OHLC in this area. Although HDT was less efficient at attracting Anopheline mosquitoes, it was highly efficient at trapping culicine mosquitoes.

Background: Whilst the human landing catch (HLC) technique is considered the 'gold standard' for estimating human-biting rates, it is labor-intensive and fraught with potential risk of exposure to infectious mosquito bites. This study evaluated the feasibility and performance of an alternative method, the human double net trap (HDNT) relative to HLC for monitoring host-seeking malaria vectors of the Anopheles minimus complex in a semi-field system (SFS). Methods: HDNT and HLC were positioned in two rooms, 30 m apart at both ends of the SFS. Two human volunteers were rotated between both traps and collected released mosquitoes (n = 100) from 6:00 pm till 6:00 am. Differences in Anopheles mosquito densities among the trapping methods were compared using a generalized linear model based on a negative binomial distribution. Results: There were 82.80% (2,136/2,580) of recaptures of wild-caught and 94.50% (2,835/3,000) of laboratory-reared mosquitoes that were molecularly identified as An. harrisoni and An. minimus, respectively. Mean density of An. harrisoni was significantly lower in HNDT (15.50 per night, 95% CI [12.48-18.52]) relative to HLC (25.32 per night (95% CI [22.28-28.36]), p < 0.001). Similarly, the mean density of a laboratory strain of An. minimus recaptured in HDNT was significantly lower (37.87 per night, 95% CI [34.62-41.11]) relative to HLC (56.40 per night, 95% CI [55.37-57.43]), p < 0.001. Relative sampling efficiency analysis showed that HLC was the more efficient trap in collecting the An. minimus complex in the SFS. Conclusion: HDNT caught proportionately fewer An. minimus complex than HLC. HDNT was not sensitive nor significantly correlated with HLC, suggesting that it is not an alternative method to HLC.
